That said, I was wondering if you remembered what type of sequencer you used for your 2b-RAD project? I’d assume an Illumina machine, but was it a Hiseq? Miseq? How many cycles did you run as well?
I’m so glad! I did about 70 samples per 50bp Illumina HiSeq 2500 lane. For PCR cycles I did a titration similar to that recommended in the Meyer’s protocol and settled on 17. Less is definitely more for cycles!
